Application of Apogee flow cytometry to the counting of CD4+ T cells in the blood of AIDS patients

Abstract : Researchers from the teaching hospital of LeDong Taike University published an article entitled " Evaluation of a flow cytometry method for CD4+ T cell enumeration based on volumetric primary CD4 gating using thermoresistant reagents ", which reported a test for AIDS. The convenience and cost of the number of CD4+ T cells in the patient's blood is compared with traditional methods, emphasizing the broad utility of the method under limited resource conditions. The article is published in the latest Journal of Immunological Methods .

HIV can attack the patient's immune system. Because of the low resistance, the human body eventually loses the opportunity to replicate immune cells, and infects other diseases, causing various diseases to co-infect and die. Therefore, in the process of clinical treatment and prevention of AIDS, real-time detection of the number of patients with CD4 + T cells will help to carry out targeted treatment work. The most common method currently used is to count CD4+ T cells on a conventional flow cytometer . However, conventional flow cytometry is too large and expensive, and it cannot meet the needs of AIDS prevention and treatment in poor areas.

In this study, the researchers evaluated the use of a heat-resistant reagent, a simple, mobile, and durable flow cytometer, the Apogee Auto 40 analyzer ( Auto40 ), for CD4+ T cell counting. The absolute number of CD4 Statistical fresh blood samples from 170 subjects in Senegal + T cells, including the 129 HIV + patients and 41 HIV - group. After a volume of CD4+ T cells were collected, the cells were stained with a commercially available reagent (Easy MoAb CD4; Bio-D, Valenzano, Italy) and analyzed using Auto40 . The results of the analysis were compared with the results of the analysis by the FACSCount system (Becton Dickinson, San Jose, USA) .

The results show that the analysis results of Apogee Auto 40 are in good agreement with the analysis results of traditional flow cytometry. Moreover , the heat-resistant reagent used in Apogee can successfully complete the counting of CD4+ T cells under the condition that the cooling system cannot be guaranteed . These advantages help to quickly and easily perform laboratory HIV testing with limited resources and configuration , effectively reducing the cost of routine HIV testing.

Apogee flow cytometer breaks through the traditional flow cytometer, has unmatched sensitivity ( <100nm ), can detect microorganisms two orders of magnitude smaller than ordinary bacteria, and is a leader in microbial detection. Only light scattering, bacteria, archaea, and most viruses can be accurately detected.
